EKSPLORASI POTENSI KAROTENOID SEBAGAI TERAPI PENDUKUNG PADA DIABETES MELLITUS TIPE 2: REVIEW ARTIKEL

Diabetes Mellitus Tipe 2 (DMT2) merupakan gangguan metabolik kronis yang ditandai oleh hiperglikemia kronis akibat resistensi insulin dan disfungsi progresif sel β pankreas. Hiperglikemia kronis dan lipotoksisitas meningkatkan produksi reactive oxygen species (ROS) yang memicu stres oksidatif, inflamasi, gangguan sinyal insulin, serta kerusakan sel β pankreas. Mengingat pentingnya peran stres oksidatif dalam patogenesis DMT2, senyawa antioksidan alami memperoleh perhatian sebagai terapi pendukung yang potensial. Tinjauan ini bertujuan untuk mengevaluasi potensi karotenoid sebagai agen terapi pendukung dalam pencegahan DMT2. Metode yang digunakan berupa literature review melalui penelusuran basis data elektronik, meliputi Google Scholar, ScienceDirect, ProQuest, dan PubMed terhadap artikel yang dipublikasikan pada tahun 2016–2026. Artikel yang dipilih mencakup artikel tinjauan, studi praklinis, dan studi klinis yang membahas efek karotenoid terhadap DMT2. Berbagai bukti ilmiah menunjukkan bahwa karotenoid, khususnya likopen, astaxanthin, β-karoten, dan lutein, memiliki aktivitas antidiabetik yang signifikan melalui berbagai mekanisme molekuler. Senyawa tersebut berperan sebagai antioksidan dengan menurunkan stres oksidatif, menekan jalur inflamasi, melindungi sel β pankreas, meningkatkan sensitivitas insulin, serta meningkatkan transpor glukosa melalui modulasi jalur sinyal PI3K/Akt dan AMPK. Secara keseluruhan, karotenoid menunjukkan potensi yang menjanjikan sebagai intervensi nutrasetika untuk mencegah dan memperlambat progresivitas DMT2. Namun, penelitian klinis berskala besar masih diperlukan untuk mengonfirmasi efektivitas jangka panjang serta aplikasinya sebagai terapi pada manusia.
