Latar belakang: Staphylococcus aureus adalah salah satu bakteri penyebab morbiditas dan mortalitas penyakit infeksi yang paling sering di seluruh dunia. Obat tradisional di Indonesia dapat dikembangkan lebih lanjut dalam upaya peningkatan pelayanan kesehatan. Tujuan penelitian lebih lanjut terkait tanaman tradisional juga sangat diperlukan mengingat penggunaan obat antibiotik untuk mengatasi bakteri Staphylococcus aureus memiliki potensi untuk menimbulkan resistensi dan hal tersebut akan berdampak pada pemilihan obat.Metode: Metode yang digunakan yaitu metode difusi Kirby-Bauer Disc Diffusion. Metode difusi cakram Kirby-Bauer merupakan metode yang paling sering digunakan untuk penentuan sensitivitas agen antimikroba dari bakteri yang diisolasi dari spesimen klinisHasil: Pada kelompok ekstrak getah biduri 100% didapatkan rata-rata zona hambat 19,13 mm, kelompok 75% didapatkan rata-rata zona hambat15,80 mm, kelompok 50% didapatkan rata-rata zona hambat adalah 9,73 mm, kelompok 25% didapatkan rata-rata zona hambat 4,77 mm, kelompok amoxicillin (K+) didapatkan rata-rata zona hambat 34,57 mm, dan pada kelompok DMSO (K-) didapatkan rata-rata zona hambat 0.00 mm.Kesimpulan: Adanya aktivitas antibakteri ekstrak getah biduri (Calotropis gigantea) terhadap pertumbuhan bakteri Staphylococcus aureus secara in vitro. Dari hasil zona hambat terendah dihasilkan oleh kelompok ekstrak getah biduri 25% sedangkan zona hambat tertinggi dihasilkan oleh kelompok ekstrak getah biduri 100%.Downloads
